dc.description.abstractLaboratóriumunkban asszisztált reprodukciós technikák (ART) számos alkalmazásával foglalkozunk. Kutatási témám a haszonállatokon végzendő preimplantációs genetikai diagnózis módszerének, protokolljának kifejlesztése. Ennek első fázisaként, kísérleteinkben a preimplantációs genetikai diagnózist kívántuk ivarmeghatározásra alkalmazni szarvasmarha (Bos primigenius taurus) és háznyúl (Oryctolagus cuniculus domestica) fajokban, egy sejt PCR alkalmazásával. A 8-16 sejtes zigóta mikromanipuláció segítségével kinyert egy blasztomerjének vizsgálatával így lehetővé válik az embrió ivarának beágyazódás előtti meghatározása, majd a diagnózis alapján az embriók ivarszelektálása is, mellyel időt és költségeket spórolva képesek vagyunk a születési ivararányt a kívánt mértékben eltolni. Nyúlban még nincs leírt egy sejt PCR-rel dolgozó PGD protokoll, mely laborunkban a nyúl méhbeli elhelyezkedés - genitoanális távolság összefüggéseinek vizsgálatához fontos lenne. Laborunkban korábban már működő szarvasmarha ivarmeghatározás módszerét és körülményeit újra beállítva pedig lehetővé válik, hogy ivarszelektált embriókat ültessünk vissza a recipiens állatokba, melynek mind a tej, mind a húsmarha tenyésztésben fontos szerepe van.hu
dc.description.abstractIn our laboratory we examine the different use of assisted reproductive techonolgy (ART). The theme of my research is to develop a method and protocol for using preimplantation genetic diagnosis on livestock animals. First we wanted to make a new protocol for sexing cattle (Bos taurus primigenius) and house rabbit (Oryctolagus cuniculus domestica) species, using single cell PCR. The preimplantation genetic diagnosis is the earliest form for the prenatal diagnostication, with that it is possible, heritable and inheritable properties and diseases to examine and on the score of there, to select the embryos before the implantation. In 1967 Edwards and Gardner made a chromatin analysis from trophectoderma cells of rabbit embryos, and predicted the gender of embryos before their transfer. This was the base to develop the method of PGD (Sexing of Live Rabbit Blastocysts, R. G. Edwards & R. L. Gardner, 1967). For human practice, PGD was developed the late 1980’s, but its potential was quickly discovered of the livestock assisted reproductive procedures. Using the selection of the pre-implantational embryos, we are able to transfer just embryos, which are possess the desired properties to the recipients, hereby, to increase the time and cost efficiency. This is especially important in case of properties, which manifest in latest life-stages, and in addition, the examined animals have a long life cycle, or they are able just one or few offspring to birth/gestate at time. These are all typify for cattle, which was one of the first livestock species on which the PGD is used. In case of cattle, the gender has a very important role in terms of the later recovery, because dairy cattle breeds, is preferred the birth of females (9:1 sex ratio is possible, in favor of females), while at beef cattle preferable to the birth of males. The develop of PGD for the case of the second observed species was important, because we want to make a negative control for Altbäcker’s experience. The hypothesis of these investigations that the male foetus in the rabbit’s uterine can effect for the female foetuses genitoanal distance, which can indicate the future agressivity and attractiveness of females. We developed SRY-based sexing protocols for rabbit and cattle, and with these, we are able to implant back into the recipients.en
